I am preparing to build a light table for registering my films with my Tri-Loc. What is the best material to use, plexi, or just go with a frosted glass?

imo: White translucent Plexi with glass over top. If you don't use glass over top, the plexi will be scratched all to crap very quickly.

What I did was butcher an old vastex flouro unit, I cut the pan down to size and had the glass cut down to match. I then stuck white sign vinyl to the glass and nopw with the bulbs on it shines through perfectly.

Look on craigslist in your area. I got mine for $100. They are out there cheap.

I made this a few years back to help register my films on the screens, works great and every film is in the same place, there's a one or two more things I need to add to it just have not gotten to it yet. Oh and I made this from plexiglass.

You may want to check out your local classifieds, craigslist etc. I was going to build but picked up a perfect size HENNING light table from the paper printing industry, there were lots in my area and I waited until the size i wanted came up . Only payed $50.00
